MASCoT utilizes intermolecular disulfide linkages to create flexible protein-protein interfaces that serve as an evolutionarily-naïve surface on which to forge diverse metal-binding sites. This strategy does not require unnatural amino acids or cofactors, benefits from an expedient workflow and is potentially generalizable to a wide range of protein monomers.
