Quantification of relative levels of AcnA in STM WT and ΔsodAB. STM WT, ΔsodAB and ΔacnAB were cultured aerobically in LB broth for 3.5, 8, 14, and 18.5 h at 37°C, cells were harvested and lysed. Proteins were separated using SDS-PAGE on 10% gels and blotted onto nitrocellulose membrane. Blots were incubated with anti-AcnA serum and goat-anti-rabbit-HRP antibody, followed by detection using an ECL kit and Chemidoc system. Blots were stripped and incubated with anti-DnaK antibody and goat-anti-mouse-HRP antibody and subsequent detection.
