Crizotinib activates Erk1/2 by inducing Ca2+ influx into the cytoplasm. (a) SPC-A1 and PC-9 cells were treated with crizotinib for the indicated amount of time, quantification of fluorescence intensity was detected by flow cytometry. (b) SPC-A1 and PC-9 cells were treated with the indicated concentration of crizotinib for 12 h, quantification of fluorescence intensity was analyzed by flow cytometry. (c) Cells were treated with TG for the indicated amount of time, after which the cells were examined for the expression of phosphorylated and total Erk1/2 by immunoblotting. (d) Cells were incubated with crizotinib in the presence or absence of EGTA for 12 h, after which they were examined for the expression of phosphorylated and total Erk1/2 by immunoblotting.
