Dasatinib decreases the translation efficiency of CHIKV RNA. (A) NHDFs infected with CHIKV (MOI = 3 PFU/cell) were untreated or treated with 10 μM dasatinib. Cytoplasmic lysates of infected cells were collected at 12 hpi and resolved through 10 to 50% linear sucrose gradients. The presence of ribosomal subunits (40S and 60S), monosomes (80S), and polysomes was monitored by continuous measurement of the absorbance (OD254) during fractionation. (B) Total RNA was prepared from each fraction, and the percentage of CHIKV RNA per fraction was determined by qRT-PCR as a percentage of the total cytoplasmic viral RNA. (C) The translation efficiency (TE) of CHIKV RNA under each condition was determined by qRT-PCR. The fold change in translation efficiency for CHIKV RNA in the presence of dasatinib is reported. The translation efficiency of the vehicle-treated control cells is set to 1 (n = 2).
