RDE (II) inactivates anti-HA IgE but not anti-HA IgG. A, HEK293T cells were transfected with pCADEST1–anti-HA IgG or anti-HA IgE and anti-HA κ. One week later, the supernatants were collected and treated with RDE (II) for 6 h. Then, the antigen-binding level of anti-HA IgE and anti-HA IgG was analyzed by competitive ELISA. B, expression level of the RDE (II)–treated antibodies in the supernatant was analyzed by quantitative ELISA coated with anti-mouse Ig. C, supernatants that were treated with RDE (II) were blotted under nonreducing conditions. They were analyzed with HRP-conjugated light chain BP.
