ZAP interacts with the Nsp9 of PRRSV. (A) Western blot of Flag-tagged PRRSV proteins and HA-tagged ZAP from whole-cell lysates (WCL) of cotransfected 293T cells and of the proteins immunoprecipitated with rabbit anti-HA. (B) Western blot showing that the ZAP-Nsp9 interaction is independent of RNA. pFlag-Nsp9- and pHA-ZAP-cotransfected cell lysates were collected and treated with RNase A, and then co-IP was performed using mouse anti-Flag. (C) Colocalization of ZAP and Nsp9. Cotransfected 293T cells were stained for Flag-Nsp9 (green) and HA-ZAP (red), with nuclei stained with the DNA-binding dye DAPI (blue). Experiments were performed in triplicate.
