DmNopp140-RGG and DmNopp140 localize to the DFCs of Xenopus oocyte nucleoli, but they fail to localize to the sphere organelles. Xenopus oocytes were injected with synthetic transcripts that encoded either GFP-DmNopp140-RGG (A–E) or GFP-DmNopp140 (F–J). (A, D, F, and I) Phase contrast images showing a few of the ∼1000 nucleoli in each oocyte nucleus. GFP-DmNopp140-RGG (B) and GFP-DmNopp140 (G) both localized within the DFC regions of the nucleoli, specifically within a subregion immediately surrounding the FCs. These FCs contained detectable amounts of the nucleolar DNA as demonstrated by DAPI staining (C and H). Some nucleoli typically contained multiple FCs and DFCs (G and H).
