CORM-2 induces the formation of a c-Src/EGFR/p47phox/Nox2 complex. (A) HPAEpiCs were transfected with scrambled, p47phox, or Nox2 siRNA and then incubated with CORM-2 for 16 h. The levels of p47phox, Nox2, HO-1, and β-actin protein were determined by western blot. (B,C) Cells were treated with CORM-2 for the indicated time intervals. The cell lysates were subjected to immunoprecipitation using an anti-Nox2 or anti-EGFR antibody. The immunoprecipitates were analyzed by western blot using an anti-c-Src, anti-EGFR, anti-Nox2, or anti-p47phox antibody. (D) HPAEpiCs were transfected with scrambled, p47phox, or Nox2 siRNA and then incubated with CORM-2 for the indicated time intervals. The levels of p47phox, c-Src, and EGFR phosphorylation were determined by western blot.
