The effect of intracellular Ca2+ chelator 1,2-Bis(2-aminophenoxy)ethane-N,N,N′,N′-tetraacetic acid tetrakis (acetoxymethyl ester) (BAPTA-AM) on intracellular events associated with MK6-83-induced apoptosis in glioma cells. (a) BAPTA-AM (10 μM) was applied 2 h before the addition of MK6-83 for 24 h in T98 and for 48 h in U251. Biparametric flow cytometric analysis was performed by Annexin V-FITC and PI staining. (b) Lysates from T98 and U251 cells pretreated with BAPTA-AM and then treated with MK6-83 for 24 h in T98 and for 72 h in U251 were separated on SDS-PAGE and probed with anti-caspase-3 Ab. Blots are representative of three separate experiments.
