IQGAP1 activates the Wnt/β-catenin signaling pathway and promotes β-catenin translocation into the nucleus. Total β-catenin, cyclin D1and c-myc protein (A) and mRNA levels (B) were assayed by western blotting and qRT-PCR, respectively, in IQGAP1 knockdown, IQGAP1 overexpression, and control groups. (C,D) Protein expression of cytoplasmic or nuclear β-catenin and its target genes (cyclin D1 and c-myc) in IQGAP1 knockdown, IQGAP1 overexpression and control groups was assayed by western blotting. (E) Immunofluorescence further revealed the change in protein expression and distribution of β-catenin by IQGAP1 knockdown. Nuclei were visualized by DAPI staining.
