Visualization of intestinal CD8+DCs. (A) To confirm the presence of intestinal CD8+ DCs, total leukocytes from rainbow trout intestine were fixed and stained with anti-CD8α (green) and anti-MHC II (red) mAbs, counterstained with DAPI (blue) and analyzed by fluorescence microscopy. Scale bars, 5 μm. (B) In order to study the location of CD8+ DCs within the tissue, cryostat sections were prepared from rainbow trout intestine, fixed and labelled with anti-CD8α (green) and anti-MHC class II (red) Abs, counterstained with DAPI (blue), and analyzed by fluorescence microscopy.
