Cytoprotective and growth-stimulatory effects of endogenous iNOS/NO in PDT-challenged U87 cells. Cells at ~65% confluence in 35-mm dishes were pre-incubated with ALA (1 mM, 30 min), and then irradiated (1 J/cm2) in the absence vs. presence of 1400W (50 μM) or cPTIO (50 μM). The viable cell fraction was then determined over 24 h of post-irradiation incubation, using the CCK-8 assay. At 24 h post-hv, some cultures were washed and proliferation of remaining live cells in 10% FBS-medium −/+ 1400W or cPTIO was tracked over an additional 24 h. Plotted values are means ± SE (n=3).
