Removal of neutrophil GAGs specifically ablates migration along a CXCL8 gradient. (A)–(F) show the collated tracks of individual migrating neutrophils (duplicate conditions) pooled from three independent experiments using different donors. In (A)–(C), neutrophils were incubated in buffer alone prior to migration, whereas in (D)–(F), cells were treated with a glycanase mixture. Gradients of fMLF, CXCL8 were established where noted, with the total number of tracked cells shown in the top right-hand corner. (G)–(I) show significant differences in velocity, directionality, and FMI of the data in (A)–(C) following tracking analysis. Error bars represent the SEM.
