Ca2+ binding pocket disruption does not influence general AC catalytic activity. MLO-Y4s were transfected with AC3 (yellow) and AC6 (green) or their corresponding mutation overexpression plasmids and treated with forskolin. Compared to vehicle controls, all specimens demonstrated an approximate 2-fold increase in cAMP production. This increase was comparable between OE plasmids and their Mut OE counterparts, as well as between AC3 and AC6 specimens. Data are normalized to vehicle controls and reported as the mean ± SEM. n = 5 for AC3 specimens, n = 6 for AC6 specimens. This experiment was performed 3 times for each group.
