SFTSV suppresses MHC-II and CD86 expression on activated macrophage. (A–C) THP-1 cells were infected with SFTSV UV-inactivated SFTSV or mock-infected at an MOI = 1. After non-specific binding being blocked by human TruStain FcX™ (BioLegend, USA), CD11b+ macrophages were analyzed for cell surface expression of HLA-DR (A), CD86 (B), and CD40 (C) by FACS using the indicated antibodies: Mock (orange line), UV-inactivated SFTSV (red line), or SFTSV (blue line). (D–F) Four-week-old male C57/BL6 mice (n = 4) were inoculated with 105 TCID50 SFTSV per mouse through intravenous injection and four mock-infected mice as controls. After non-specific binding being blocked by mouse Fc Block™ (BD Biosciences, USA), mouse splenic CD11b+ F4/80+ macrophages were analyzed for cell surface expression of MHC-II (D), CD86 (E), and CD40 (F) by FACS using the indicated antibodies: Mock (orange line), 5d SFTSV (blue line), or 9d SFTSV (red line).
