LdCen^−/− immunization induced Th17 along with the inhibition of anti-inflammatory cytokines compared with LdWT infection in splenocytes. LdCen^−/−-immunized or LdWT-infected mice were sacrificed at 2 and 5 wk postinfection. Splenocytes were collected and cultured in presence of Leishmania Ag. Culture supernatants were collected and concentrations of cytokines IL-17A, IL-17F, IL-6, IL-1β, IL-22, IL-23, IL-27, IL-10 (A–H) were measured by the multiplex or single plex (IL-27) mouse cytokine ELISA kit as described in the Materials and Methods section.
