Whole blood samples and PBMCs were obtained from healthy individuals. Expression of surface FPN1 (n = 10), TfR1 (n = 10), DMT1 (n = 7), ZIP14 (n = 7), CD172a (n = 6), TIM4 (n = 6), and intracellular ferritin (n = 4) in monocyte subsets was measured by flow cytometry. Blood monocyte subpopulations were defined as presented in Supplemental Figure 2A (red: classical; gray: intermediate; blue: nonclassical monocytes). (A) Representative signal histograms (open histograms: isotype; tinted histograms: specific antibody). (B) In graphs with ΔMFI values, each point represents 1 measurement, bars denote mean, and error bars represent SEM. Statistical significance was assessed with first-order linear models. Estimates for protein expression in classical monocytes and for differences in expression between subsets are presented with 95% CI. Estimate P values were calculated with 2-tailed t test. ANOVA statistics for each marker are shown in Supplemental Table 3.
