Inhibition of DENV2 replicon replication by curcuminoids (A) BHK-21/DENV2 replicon expressing cells (~5 ×103 in 100 μl) were seeded onto 96-well plates and incubated at 37oC for 24 h in a humidified incubator with 5% CO2. Compounds were added at 0, 0.78, 1.56, 3.12, 6.25, 12.5, 25, 50,100 and 200 μM in 1% DMSO in triplicate wells. Cells were incubated at 37 °C for 24 h, washed, lysed, and Rluc activi ties were measured. The results were confirmed by two independent experiments. (B) Cytotoxicity was measured in the same replicon expressing cells treated with the various concentrations of compounds at 24 h using the Cell Counting Kit −8 following the manufacturer’s instructions.
