Mouse PXR agonists fail to evoke caspase-1 activation or IL-1β release in peritoneal macrophages isolated from PXR/ mice. The PXR is expressed in LPS-pulsed peritoneal macrophages (A) and is required for PCN-induced caspase-1 activation [as assessed by detecting the cleaved caspase-1 p20 subunit in the supernatant (B and C)] and subsequent IL-1β release (D), measured by enzyme-linked immunosorbent assay.
