A phosphorylation-mimic Hjc mutant S34E completely lost the HJ DNA cleavage activity. Ten nanomolar wild type Hjc or its mutant was incubated with HJ at 65°C in 20 μl reactions (see the Materials and Methods). The samples were mixed with loading buffer, boiled for 10 min, and analyzed on a 15% denatured PAGE gel containing 7 M urea. The position of a 34-nt oligonucleotide (Supplementary Table S2) is indicated at the left side of the gel. The experiments were performed at least for three times. A representative image is shown as the left panel and the quantitation is shown as the right panel. Error bars indicate standard deviation.
