Measurement of mitochondrial permeability using JC-1 dye following exposure to 1 μM CdCl2 in HPNE and AsPC-1 cells. Fluorescence intensity was measured at 535ex/595em to measure JC-1 aggregates (healthy cells; green) and then a second reading using 485ex/535em which measures JC-1 monomers (apoptotic/unhealthy cells; red). The ratio of green/red represents a relative number of cells which are 'healthy'. There was a significant effect of treatment (F1,16=7.349; P=0.0154) with reductions in the ratio following exposure to 1 μM CdCl2 in both groups. Individual comparisons to control values did not reveal any difference in the fluorescent ratio in either cell line. Data represent the means ± SEM of 5 assays (n=5) performed in duplicate.
