Formation of heat-labile, SDS-resistant ternary complexes with wild-type and mutant forms of VAMP-2. Combinations of soluble wild-type or mutant VAMP-2, syntaxin 1A, and SNAP-25 (2 μm each) were incubated for 16 hr at 4°C. After SDS-PAGE sample buffer was added, samples were divided into two aliquots and either incubated at 37°C (a) or boiled (b). After SDS-PAGE and transfer to nitrocellulose, multimeric complexes were detected by using an antibody against syntaxin 1A and visualized by autoradiography.
