Cell specificity of CGS inhibition of CT/CGRP enhancer activity. TK reporter genes containing the H/O enhancer (solid box) or the H/O+A mutant enhancer were transfected into HeLa cells expressing the 5-HT1B receptor, treated with 10 μm CGS or vehicle for 16 hr, and then assayed for luciferase activity. The data in each experiment were normalized to the TK–luciferase reporter activity in the presence (+) or absence (−) of CGS and expressed as the fold change relative to TK, the mean for which was set at one. The average activities for CGS-treated and untreated TK-transfected cells were 3200 and 1600 light units per 20 μg of protein, respectively. The means and SE from three independent experiments with duplicate samples are shown.
