Identification of theTr^J genotype. TheTr^J mutation introduces a novelBanI restriction site in the pmp22sequence (A). A 103 bp fragment of thepmp22 gene, including the mutated site, was amplified by PCR. BanI restriction enzyme untreated (−) and treated (+) samples were analyzed on an ethidium bromide-stained 6% acrylamide gel (B). Homozygous (Tr^J/Tr^J) and heterozygous (Tr^J/+)Tr^J animals are identified by the cleavage of the 103 bp PCR-amplified fragment into two smaller pieces (70 and 33 bp). The numbers on the left of the gel indicate DNA fragment size in base pairs.
