Anisomycin blocks PD-induced increases in the expression of BDNF transcripts containing exons I and II but not transcripts containing exons III and IV. Dark-field photomicrographs showing labeling of stratum granulosum (sg) with cRNAs for BDNF exon I- (AC), exon II- (D–F), exon III- (GI), and exon IV- (JL) containing transcripts in sections through hippocampus of a control rat (A,D, G, J), a rat killed 2 hr after a PD applied alone (B,E, H, K), and a rat killed 2 hr after a PD applied in the presence of anisomycin (C, F, I,L). Note that a PD applied alone increased hybridization of all four transcript cRNAs in stratum granulosum; anisomycin cotreatment virtually eliminated the PD-induced increase in exon I cRNA (C) and exon II cRNA (F) labeling but did not appear to attenuate the PD-induced increase in exon III (I) or exon IV (L) cRNA labeling. Scale bar, 200 μm.
