Individual PDOTS were prepared from resected patient tumors and incubated for 3 days with vehicle or compound 6 (0.5 nM in culture medium). Cells were labeled with antibodies to detect CD3 (pan-T cells), CD4 (T helper cells), CD44 (effector-memory cells), and IFNγ (marker of T cell activation) then analyzed by flow cytometry. The percentage of cells expressing the indicated marker in vehicle treated samples was set to 1 and the fold change in the presence of compound 6 determined.
