Primary mouse pancreatic acini and AR42J cells were infected with adenoviral vectors to assess NFAT and NF-κB associated luciferase activities, and then treated with 100 nM CCK in the presence or absence of CM4620. Luciferase activity was measured 4.5 h after CCK treatment and expressed as relative luciferase units (RLU) per mg of total protein. As expected, CCK induced marked NFAT-luciferase activity in mouse acini (A) and AR42J cells (B), and this effect was completely blocked by CM4620. (C) CCK also induced NF-κB activation in AR42J cells, and this effect was significantly inhibited by CM4620. Graphs show mean ± SD; n=3–4 independent experiments.
