Acute pancreatitis was induced in male Sprague-Dawley rats by intraperitoneal (IP) administration of cerulein (Cer; 50 μg/kg; 4 hourly injections); control mice received saline as vehicle control. In a subset of rats, the Orai/Stiml inhibitor CM4620 was administered at the indicated doses as a 4-h continuous intravenous (IV) infusion into the jugular vein. The IV infusion was initiated 30 min after the first cerulein injection (A, therapeutic administration of CM4620) or 2 h before the first cerulein injection (B, prophylactic administration of CM4620). Animals remained conscious during the whole procedure. Blood and tissue samples were collected post-mortem at the indicated times.
