Inhibition of the miRNA processing enzyme, DROSHA, enriches endometriotic lesion expression of pri-miR-144-3p but not pri-miR-451a. The endometriotic epithelial cell line, 12Z was transfected with either a non-targeting siRNA (NT; negative control) or DROSHA siRNA. Twenty-four and forty-eight hours after transfection, DROSHA mRNA (A) and protein (B) expression were determined as was pri-miR-144-3p (C) and pri-miR-451a (data not shown). Pri-miR-451a was not detected in any of the 12Z cell samples by qRT-PCR. Different letters indicate statistical significance (P < 0.01) among the means by one-way ANOVA followed by Bonferroni post-hoc analysis (planned comparisons).
