Membrane vesicles were incubated with 50 μM p-glucocoumaryl alcohol in the presence (●) or absence (△) of 5 mM Mg/ATP. (B) Negative control for p-glucocoumaryl alcohol uptake. Membrane vesicles of hybrid poplar were incubated with 50 μM p-glucocoumaryl alcohol for 20 min either with 5 mM Mg/ATP, with 5 mM Mg/ADP (+ADP), with 5 mM Mg/AMP (+AMP), or without Mg/ATP (−ATP). “Boiled” refers to a microsomal fraction heat-denatured for 10 min. (C) p-Glucocoumaryl alcohol uptake shows saturation kinetics. Membrane fractions of hybrid poplar were incubated for 5 min in the presence of 5 mM Mg/ATP and each concentration of p-glucocoumaryl alcohol. Inset shows Hanes–Woolf plots.
