Flow Cytometry Binding of Aptamers

(A) Comparison of the binding profiles of aptamers H02 (left) and B03 (center) on CHO-B2 cells (gray lines) and CHO-B2 α5+ cells (black lines) at 4°C. Profiles without aptamer labeling are shown on the right. (B) Titration of aptamer H02 resulted in determination of the equilibrium affinity parameter K_D for the interaction between U87MG α5+ cells and aptamers H02 (277.8 ± 51.8 nM). Cy5-aptamer H02 at concentrations of 0.15, 0.3, 0.6, 1.25, 2.5, and 5 μM was incubated on ice with a constant amount of cells and analyzed by flow cytometry.
