Notes: (A) Diagrams are shown for the binding of hsa-miR-1270 on two DNA sites on 3′-UTR of human WT WT1. The sites were also MU to void hsa-miR-1270 binding. (B) HEK293T cells were transduced with lentiviruses of miR-MC or miR-1270M. Then, they were secondarily transfected with luciferase plasmids of WT1-WT or WT1-MU, followed by a dual-luciferase reporter assay (*P<0.05). (C) In lentiviral-transduced LN-18 and A172 cells, we applied quantitative real-time PCR to examine endogenous WT1 expressions (*P<0.05). (D) In lentiviral-transduced LN-18 and A172 cells, we applied Western blot to examine WT1 protein expressions.
