Alterations of sphingolipid concentrations during C. rodentium infection. C57BL/6 mice were either left untreated (WT) or pre-treated with 180 mg/l amitriptyline in drinking water 2 weeks prior to infection (WT/Ami) and during the course of infection. Mice were orally gavaged with PBS or with ~2 × 109 colony forming units (CFUs) of C. rodentium. At indicated days post (dp) infection colons were excised. In rectal colonic tissue, the (A,C) sphingomyelin and (B,D) ceramide contents were analyzed using rapid resolution liquid chromatography/mass spectrometry. Data are presented as the concentration of ceramide and sphingomyelin in pmol/mg protein. All data are presented as mean ± SEM. Statistics were performed using the parametric one-way ANOVA test with Tukey's multiple comparison test (*p < 0.05; **p < 0.01).
