Acute application of a α5-NAM rescues NMDAR-dependent synaptic plasticity. The fEPSP slope was significantly less elevated in Ts65Dn slices (110.1 ± 3.1%, n = 19; red) compared with WT littermates (127.1 ± 7.2%, n = 14; p < 0.05, two-sample t test; black) at 80 min after induction. C, Application of the α5-NAM (0.3 μm) enhanced LTP in Ts65Dn mice up to normal levels. The potentiation of the fEPSP slope in Ts65Dn slices was increased in the presence of the α5-NAM (122.9 ± 6.2%, n = 11; blue) relative to control conditions (p < 0.05; two-sample t test), and comparable with WT slices in control condition (127.1 ± 7.2%, n = 14; black, same experiments as in B).
