miR-548b suppresses the PI3K–AKT signaling cascade in hepatocellular carcinoma (HCC) cells by downregulating HMGB1. Hep3B cells overexpressing miR-548b were next transfected with pCMV-HMGB1 or pCMV. MiR-548b inhibitor–transfected SK-HEP-1 cells were treated with si-HMGB1 or si-negative control (NC). (A) Hep3B cells were transfected with pCMV-HMGB1 or pCMV. Western blot analysis was used to determine the HMGB1 protein expression. *P<0.05 as compared with pCMV. (B) Seventy-two hours after transfection, Western blotting was carried out to measure p-PI3K, PI3K, p-AKT, and AKT protein levels. GAPDH served as a loading control.
