Overall workflow of immune repertoire mining for affinity engineering. Repertoire sequencing is performed in parallel and from tissues from the same mice used for hybridoma development. Immune repertoire is performed immediately after mAb characterization and sequencing. Initial affinity screening is done with hybrid IgG composed of a mAb and a variant chain. Combination of variant chains after initial affinity screening is not shown for simplicity.
