PpIX activates hTRPA1 and hTRPV1. A, In hTRPA1-expressing HEK293t cells PpIX (PpIX, 5 min) evoked a concentration-dependent increase in [Ca2+]i. B, Similarly, hTRPV1-expressing cells challenged with PpIX showed concentration-dependent calcium increases. C, In hTRPA1-expressing cells, PpIX increased the UV response compared with untreated hTRPA1-expressing cells or HEK293t cells. When the 340/380 nm illumination (10 ms each; 1 Hz) within the first minute of PpIX application was omitted (gray box), a direct effect on intracellular calcium was observed for PpIX 1 nm, but not for PpIX 0.1 nm.
