Inhibition of ASAH1 does arrest PGCCs and functions as a neosis inhibitor. A: PPC1 cells with scrambled or ASAH1-targeting shRNA were exposed to 5 nM docetaxel or 8 Gy radiation to form PGCCs. At 48 h, the PGCCs were allowed to recover followed by crystal violet staining with one triplicate set shown. B: PPC1 cells maintained in normal medium or in 5 μM of LCL-521 were treated with 5 nM docetaxel or exposed to 8 Gy radiation to form PGCCs. Cultures were allowed to recover for 3–6 days, n = 4. Top panels represent quantification of staining (***P < 0.001) and bottom panels shows representative images.
