(a) 1 μg HA or Flag tagged GBPs were co-expressed with increasing amounts of 6xMyc tagged IpaH9.8 in HEK293T cells as indicated. The cell lysates were analyzed by immunoblotting with indicated antibodies. The GBP3, GBP5, and GBP7 mutants are more susceptible to IpaH9.8 mediated degradation compared to the wild-type proteins. The data are representatives of three independent experiments. (b) GBP3 and GBP7 mutants display enhanced interactions with IpaH9.8-C337A. HA or Flag tagged GBPs were co-expressed with Flag or Myc tagged IpaH-C337A in HEK293T cells. Immunoprecipitation was performed using the Flag-M2 beads from the cell lysates, and associated proteins were analyzed by immunoblotting.
