The effect of TRPV4 antagonist HC and TRPV4-siRNA on the expression or activity of antioxidant enzymes in H9C2 cells exposed to H/R. The mRNA expression levels of HO-1, NQO1, SOD1, SOD2, SOD3, CAT, GST, GCSH, and GSH-Px1 were detected by real-time PCR (a–i). The protein levels of SOD1, SOD2, CAT, and GSH-Px1 were examined by western blot (j–n). β-Actin was used as an internal control. The activity of SOD (o), GSH-Px (p), and CAT (q) were determined using an ELISA. Values are presented as the mean ± SEM; n = 3-6/group. We used a one-way ANOVA followed by a Bonferroni test. p < 0.05, ∗∗ p < 0.01, and ∗∗∗ p < 0.001 vs. N; # p < 0.05 and ### p < 0.001 vs. H/R; and $ p < 0.05 and $$ p < 0.01 vs. H/R+Con siRNA.
