The number of mitotic events is increased in the VZ and SVZ of 16p11.2del mice during early and mid-neurogenesis. a, IHC of E14.5 coronal sections incubated with anti-PH3 (red). b, The number of mitotic cells was determined by counting every mitotic (PH3+) cell at the VZ and in the SVZ on three consecutive sections at E12.5 and E14.5. At E12.5, number of ventricular mitosis is increased in the 16p11del mice compared with WT littermate controls (p = 0.0173) but is not significantly elevated in the SVZ (p = 0.2654) (nWT = 8; nDel = 7). At mid-neurogenesis compared with WT, the number of mitotic cells in the 16p11.2del embryos is increased in the VZ (p = 0.08) and the SVZ (*p = 0.019), but not the ganglionic eminences (GE) (nwt = 4; ndel = 5) at E14.5. c, To confirm that increased mitosis in the SVZ is due to dividing IPCs, E14.5 cortices were stained with anti-Tbr2 (green) and PH3 (red). d, The number of Tbr2+/PH3+ IPCs is increased by 37% in the 16p11.2del mice compared with WT littermate controls at E14.5 (*p = 0.0115; nwt = 4; ndel = 3). *p < 0.05.
