Characterization of Aptamers for Cell Specificity

(A) The overall conclusion from three independent flow-cytometric analyses of ZUCH-1, ZOKT-2, ZUCH-3, ZUCH-4, and ZUCH-5 for cell specificity using Jurkat.E6 and three negative control cells: BJAB, MOLT-3, and Toledo. Percent fluorescent intensity values on the y axis were determined by normalizing against a random control sequence using the equation ((aptamerrandom/random)×100 (one-way ANOVA using Tukey’s multiple comparisons test performed on GraphPad Prism to obtain statistical significance: ***p = 0.0001; ****p < 0.0001). (B) Cross-competition experiments among ZOKT-2, ZUCH-3, ZUCH-4, and ZUCH-5, using unlabeled ZUCH-1 as a competitor against Jurkat.E6 cells, demonstrating that all five aptamers compete toward the same target on Jurkat cells.
