Impairment of autophagolysosome formation and late endosome processing. (A) Immunostaining for ubiquitin (green) and LC3 (magenta) revealed accumulation of ubiquitinated proteins colocalizing with the LC3 puncta in Vps34^fl/fl;Pcp2-Cre retinas. (B) Immunostaining for lysosomal marker LAMP1 (green) and LC3 (magenta) showed an accumulation of LAMP1-positive puncta, which did not colocalize with LC3, indicative of failure of autophagosome-lysosome fusion. (C) Late endosome marker Rab7 immunostaining (magenta) also indicated an accumulation of Rab7-positive structures in Vps34^fl/fl;Pcp2-Cre retinas.
