OXT and OXTGly directly stimulate insulin and glucagon secretion from primary islets. (A) Isolated mouse islets were treated with OXT and OXTGly for 2 h in the presence of 11 mM glucose. OXT and OXTGly dose-dependently increased insulin secretion into culture medium compared with vehicle treatment. (B) Isolated human islets were treated with OXT and OXTGly for 2 h in the presence of 3 mM or 11 mM glucose. Insulin secreted into culture supernatant was quantified. (C) Isolated human islets treated with OXT and OXTGly for 2 h in the presence of either 3 mM glucose as control or 11 mM glucose. Glucagon secreted into culture supernatant was quantified. *P < 0.05, **P < 0.01, ***P < 0.001, ****P < 0.0001, one-way ANOVA followed by a Dunnett multiple comparisons test against the vehicle control group.
