As seen in (a) there is no labeling when protein is not applied to the slide. In (b), the appearance of donor signal is seen when protein with single cysteine labeled with fluorophores is applied to the slide. In (c), the appearance of FRET is seen when protein with two cysteine labeling sites labeled with fluorophores is applied to the slide.
