Effect of Rab8 on DHPG-stimulated Ca2+ release in hippocampal neurons. Fluorescent intensity analysis of Ca2+ release as represented by the fluorescent calcium indicator fluo-4 AM. DIV7–10 primary mouse hippocampal neurons were transiently transfected with 10 μg of plasmid cDNA encoding either pEGFP (control) or GFP-Rab8. Twenty-four hours after transfection, neurons were imaged to identify transfected cells, after which they were loaded with 5 μm of the calcium indicator fluo-4 AM for 30 min at room temperature, and the same field of view was imaged for calcium release with the addition of 100 μm DHPG followed by 10 μm ionomycin (A) or 100 μm carbachol (B). Data represent the mean ± SD of three independent experiments.
