The actin florescence intensity. Human sperm (1 × 107 cells per ml) were incubated in Ham's F-10 capacitation medium for 160 min (Control). Next, 10 μmol l−1 EPAC specific inhibitor 09 (ESI09) was added for an additional 20 min. Then TG (3 μmol l− 1), polyphosphoinositide-binding-peptide (PBP10; 1 μmol l−1), N-[2-(p-bromocinnamylamino)ethyl]-5isoquinolinesulfonamide (H89; 50 μmol l−1), 8-(4-Chlorophenylthio)-2′-O-methuladenosine-3′,5′-cyclic (8pCPT; 0.1 μmol l−1] were added for an additional 1 h.
