Glutamine synthetase binds to ZNRF1. A, Proteins that bound to His-ZNRF1 from mouse brain extract with or without a Zn2+ chelator, TPEN, were purified and separated by SDS-PAGE. Note that ∼40 kDa ZNRF1-binding protein was detected only in the absence of zinc chelator. B, Neuro-2a cells were transfected with expression plasmids for GS-Myc and/or ZNRF1-Flag, and the cell lysate was subjected to immunoprecipitation. Immunoblot analysis showed that anti-Myc immunoprecipitates contained ZNRF1-Flag and anti-Flag immunoprecipitates contained GS-Myc.
