Movement‐defective Ourmia melon virus (OuMV) constructs do not form protruding tubular structures in protoplasts. Protoplasts were prepared from Nicotiana benthamiana and Arabidopsis thaliana leaves agroinfiltrated with pGC‐RNA1, pGC‐RNA3 and (alternatively) pGC‐GFP:MPwt, pGC‐GFP:MP98, pGC‐GFP:MP150 or pGC‐GFP:MP169. Fluorescence was assayed by confocal microscopy. Protoplasts were prepared at 3 days post‐inoculation (dpi). For each host–OuMV combination, at least 100 protoplasts were assayed. Scale bars, 10 µm.
