UV‐crosslinking competition analysis of the interaction between the helicase‐like domain of Bamboo mosaic virus (BaMV) replicase and Ba‐77 with unlabelled competitors. The purified helicase‐like domain (1 nmol) was pre‐incubated with unlabelled RNAs with the mass (A) or molar (B) excess amount as indicated above each lane. (A) Competitors are the unlabelled double‐stranded RNA (polyIC) and single‐stranded RNAs (polyA, polyG, polyU and polyC). (B) Competitors are the unlabelled Ba‐39 (the small stem‐loop), Ba‐77/Δ31 (the large stem‐loop), Ba‐77 and yeast tRNA. After UV‐crosslinking for 10 min, samples were digested with RNase A and then loaded onto a 12% sodium dodecylsulphate‐polyacrylamide gel. All UV‐crosslinking data presented are the average and error bars of at least three independent reactions.
